The melanocortin 4 receptor (MC4R) regulates the appetite of mammals by stimulating food intake and preventing energy expenditure. The promoter region of the human MC4R contains a polymorphic site within the transcriptional start site. Here, we investigated whether this polymorphism influences energy homeostasis. We used a glucose clamp, an euglycaemic-hyperinsulinemic clamp, and a mixed meal challenge test. The polymorphism was not associated with a change in body weight or the hyperinsulinemic response to the glucose or mixed meal challenges. The level of the MC4R transcript was not influenced by the polymorphism. We did, however, find a significant association with the body mass index in patients homozygous for the C allele. Our data indicate that this variant does not contribute to the regulation of weight homeostasis.Metformin improves insulin resistance and endoplasmic reticulum stress in the liver of a high fat diet fed mouse model. Metformin, an orally administered antidiabetic drug, has been shown to attenuate hepatic steatosis and inflammation and to enhance insulin sensitivity in the peripheral tissues. However, the effects of metformin on hepatic steatosis and inflammation and on the molecular mechanisms of its anti-inflammatory activity have not been fully explored. In the present study, we observed the effects of metformin on the livers of mice fed a high fat diet (HFD) for 14 weeks. Metformin (250 mg/kg, orally) decreased blood glucose and triglyceride levels in HFD-fed mice. Metformin decreased the plasma levels of high-sensitivity C-reactive protein, interleukin (IL)-6 and tumor necrosis factor-α (TNF-α) and increased the plasma adiponectin level in HFD-fed mice. Although metformin did not affect the plasma levels of IL-6 and TNF-α in HFD-fed mice, it inhibited the expression of IL-6 and TNF-α mRNAs in the liver. Metformin decreased the levels of mRNA of proteins associated with endoplasmic reticulum (ER) stress, inositol-requiring enzyme 1 alpha, and activating transcription factor 6 alpha and increased the expression of immunoglobulin-
